喉癌组织中VEGF mRNA及其蛋白表达与颈淋巴结转移的关系

摘要 目的研究喉鳞癌中血管内皮生长因子(vascular endothelial growth factor,VEGF)mRNA及其蛋白表达与颈淋巴结转移的关系。方法应用半定量逆转录-聚合酶链式反应(semi-RT-PCR)技术和免疫组织化学方法,检测60例喉癌中VEGF mRNA及其蛋白的表达情况。结果声门上型伴颈淋巴结转移的喉癌(第1组)的VEGF mRNA相对吸光度(A值)为0.676±0.032,声门上型不伴颈淋巴结转移的喉癌(第2组)A值为0.465±0.024,声门型不伴颈淋巴结转移喉癌(第3组)A值为0.273±0.030,无颈淋巴结转移的喉癌(第2加第3组)A值为0.369±0.113;第1组A值分别与后3组相比较,结果均有统计学意义(P均<0.01)。第1组中VEGF蛋白表达的阳性率为65.0%,第2组为20.0%,第3组为25.0%,第2加第3组总计为22.5%,第1组阳性率分别与后3组相比较,均有显著性差异(P<0.05,P<0.01)。结论在基因和蛋白水平上,VEGF的表达与喉癌的颈淋巴结转移均密切相关,VEGF基因有可能成为预测和早期诊断喉癌转移的生物学指标,并有可能成为抗血管生成治疗的靶点,或监测疗效的指标。 Objective To study the relations between the expression of the vascular endothelial growth factor (VEGF) mRNA, protein and cervical lymph nodes metastasis in laryngeal squamous cell carcinoma(LSCC). Methods The expression of VEGF mRNA and protein in 60 cases with LSCC were detected by semi-RT-PCR technique and immunohistochemistry. Results The VEGF mRNA A of supraglottic LSCC with (the first group) and without (the second group) cervical lymph nodes metastasis were 0. 676 ± 0. 032 and 0. 465 ± 0. 024 respectively; the VEGF mRNA A of glottic LSCC without cervical lymph nodes metastasis (the third group) was 0. 273 ± 0. 030 ; LSCC without cervical lymph nodes metastasis ( the fourth group, that the second group plus the third group ) was 0. 369 ± 0. 113 ( all P 〈 0. 01 ). The positive rate of the VEGF protein expression in the first group was 65.0%, in the second group was 20. 0%, in the third group was 25.0% and in the fourth group was 22. 5%. The differences were statistically significant among the first group and the others ( all P 〈0. 05 ). Conclusions The expression of gene and protein of VEGF is related to cervical lymph node metastasis of LSCC. The VEGF mRNA would become a biologic index to prognose LSCC and a target for anti-angiogenic therapy.
