摘要
目的观察四物汤配方颗粒对骨髓抑制小鼠造血功能的影响。方法50只小鼠随机分成5组,正常组、照射处理组及3个不同剂量组。应用流式细胞术(FCM)检测小鼠骨髓细胞周期,采用原位杂交技术测定骨髓细胞凋亡基因Bcl-2 mRNA和BaxmRNA表达。结果四物汤配方颗粒各组均能明显促进骨髓G0/G1期细胞向S期细胞以及S期细胞向G2/M期细胞的转化,增殖指数(PI)明显升高。提高骨髓细胞中Bcl-2 mRNA的表达,降低BaxmRNA的表达。结论四物汤配方颗粒通过促进骨髓抑制小鼠细胞周期的转化及抑制骨髓细胞凋亡,而达到促进造血功能的目的。
Objective To investigate the effect of abstracted formula granule of siwu drugs on hematogenesis function in bone marrow depression mice and to explore its mechanism of hematopoietic regulation. Methods 50 mice were divided into 5 groups randomly : a normal group, a control group and 3 dose groups. The effect of the formula granule on the cell cycle of BMCs in bone marrow depression mice was explored by flowcytometry ( FCM), and the influence of the Bcl-2 mRNA and Bax mRNA expression were determined by in situ hybridization. Results It was found that all dose groups could obviously promote G0/G1-S phase and S-G2/M phase transition and increase the proliferation indexes. It could also increase the expression of Bcl-2 mRNA and decrease the expression of Bax mRNA. Conclusion The results suggested that formula granule of Siwu drugs may promote the bone marrow cell cycle transition, inhibit apoptosis and recover the hematopoietic function in bone marrow depression mice.
