摘要
应用逆转录聚合酶链反应(RT_PCR)方法快速克隆人甲状腺过氧化物酶(hTPO)抗原决定簇基因1702~2622(编号相对于起始密码,编码AA568~AA874)。将此片段重组入PUC18质粒,并用PCR_双链DNA(dsDNA)方法测定全部顺序。结果表明确实克隆到hTPO(1702~2622)区的基因片段,有两个单碱基。
The human thyroid peroxidase (hTPO) epitopes gene (1702_2622, encoding AA568_AA874) was amplified by RT_PCR and inserted into PUC18 vector for cloning in the present study. The cloned gene was sequenced with dsDNA Cycle Sequencing System. The identity of the cloned hTPO epitopes gene was demonstrated. There were 2 bases different from the cDNA sequence published by Kimura. These changes did not interfere with translation of the protein.
