摘要
为研究压力对筛板细胞形态和糖蛋白合成的影响,对筛板细胞进行体外培养并施与一定压力,相差显微镜下观察筛板细胞形态学改变,并采用显微荧光光度计对筛板细胞合成层粘蛋白(LaminaLN)及纤维连接蛋白(fibrinectionFN)进行定量测定。结果在静压力(50mmHg)下,筛板细胞形态变扁,随加压时间延长,胞浆内空泡增多,线粒体肿胀、部分空泡化;胞浆内可见髓鞘样结构;加压3天的筛板细胞合成LN及FN分别为42.2±4.67、63.65±11.20荧光强度(AU),与对照组比较,差异呈显著性(P<0.05),加压5天的筛板细胞合成LN及FN分别为36.65±8.45、60.90±11.91AU,与对照组比较仍有不同程度增高。显示压力能促使筛板细胞形态异常和糖蛋白合成增加。
To study the influence of pressure on glycoprotein synethesis of human lamina cribrosa cell (HLC) in vitro. HLCs were cultured in vitro and submitted to elevated hydrostatic pressure, synethesis of fibronection (FN) and lamina (LN) were measured with fluorescence spectrophotometer.Result:Under a hydrostatic pressure,HLCs became flat and elongated,Vacuoles in the cytoplasm increased,mitchondria were swollen,and some turned into vacuoles,and myelinfigures were found in cytoplasm.When exposed to elevated pressure for 3 days,the amounts of synthesized FN and LN wrer 63 65±11 20,42 20±4 67 arbitrary unit (AU) by fluoremetry respectively.Compared to controls,they were statistically significantly (P<0 05)increased;and after 5 days of pressure esposure,the amounts were 60 90±11 30,36 65±8 45AU,e.g,still increased.Conclusion: Under elevated hydrostatic pressure,HLCs showed obvious morphological change and increased synethesis of FN and LN.
