脂质体介导pcDNA3.1-HGF瞬时转染小鼠MSCs及转染效果检测 被引量:1

摘要 肝细胞生长因子(HGF)对各种组织都具有促分裂、促迁移、促细胞形态发生的作用,尤其是肾脏、肺和肠道.间充质干细胞(MSC)是一种具有多向分化潜能的干细胞,同时还具有相对的定位归巢以及免疫调节作用,由于MSC具有较低的免疫原性,可作为良好的基因载体.为联合应用HGF和MSC的作用,本实验在分离培养小鼠MSC并鉴定后;应用脂质体介导法将pcDNA3.1-HGF瞬时转染MSC;采用半定量RT-PCR和ELISA法分别鉴定HGF的表达情况.结果表明:转染试剂对MSC的活性无影响,转染细胞形态及定向分化特性未发生改变;HGF在转染后48 h达到最高表达量,随时间推移逐渐减低,在10 d时仍略高于对照.脂质体介导pcDNA3.1-HGF瞬时转染小鼠MSC后,HGF在转染后10 d内升高表达. Hepatocyte growth factor (HGF) has been applied in tissue repair. In this study, bone marrow-derived mesenchymal stem cells (MSC) were isolated, expanded, characterized, and transfected with plasmid pcDNA3. 1-HGF. Reversed transcription polymerase chain reaction (RT-PCR) and ELISA analysis of mRNA and protein showed clear HGF expression at high level, which could he maintained for about 10 days. It is therefore concluded that transient HGF gene transfection in MSC could be done with the liposome method.
