摘要
目的:完善区分肌纤维类型的异染ATPase染色方法,为相关的实验研究提供借鉴和参考。结果发现:染色方案一可显示4种肌纤维类型:I、IIA、liB和IIC。大鼠骨骼肌在预孵育液pH值4.35左右时,出现最佳染色结果;而蟾蜍骨骼肌在预孵育液pH值4.40~4.70都出现了较佳的染色结果。染色方案二只能区分3种肌纤维类型:I、IIA和IIB。出现较佳结果时,大鼠和蟾蜍预孵育液的pH值也不同。结论:异染腺苷三磷酸酶法区分骨骼肌纤维类型,染色方案一可区分四种肌纤维类型,而染色方案二可区分三种肌纤维类型;出现最佳染色结果时,大鼠和蟾蜍骨骼肌的染色条件稍有不同,存在种属差异。
Objective: To improve the ATPase stains method for identification of skeletal muscle fiber types and supply reference for some experiments concerned. Result: In Protocol One, four skeletal muscle fiber types (types I, IIA, IIB, and IIC) can be identified. Optimal staining of rats skeletal muscle is shown when pH of preincubation solution is about 4.35, but it of toads is 4.40 to 4.70. Only three skeletal muscle fiber types (types I, IIA, and IIB) can be shown using the substituent reagents in Protocol Two, and the pH of preincubation solution is different between rats and toads when optimal resuit occur. Conclusion: By use of metachromatic dye-ATPase method to identify skeletal muscle fiber types, four skeletal muscle fiber types can be identified using Protocol One; three skeletal muscle fiber types can be shown in protocol Two. There is difference of optimal staining conditions between rats and toads and the difference of species.
