摘要
为了解利用母血循环中胎儿单拷贝基因的灵敏度并初步探索其临床应用价值,本研究根据人类SRY基因保守顺序设计了内、外两对引物,在明确该检测灵敏度的基础上,对15例孕妇血中胎儿SRY基因进行了扩增,结果表明这一检测程序的灵敏度达到1/2×10^7(男性DNA/女性DNA)15例母血胎儿细胞原检出率与胎儿实验性别的符合率为93.3%。本实验采取的套式PCRT技术大大提高了检测的特异性和灵敏度。
To investigate the sensitivity of detection of fetal single copy gene in maternal circulation, the human Y chromosome - specific SRY gene was detected in maternal peripheral blood of 6 pregnants bearing male fetus. Protocols used were capable of making positive i-dentification of SRY gene singles in a mixture containing less than 1ug male DNA and 2 x 107female DNA. These results suggest that identification of fetal single copy gene in the maternal circulation seems rea,dily feasible with properly designed and executed polymerase chain reaction.
