摘要
目的探讨高糖对人血管平滑肌细胞清道夫受体CD36表达的影响及对肌源性泡沫细胞产生的作用。方法体外用不同浓度的葡萄糖(5、11、20及30mmol/L)干预人脐血管平滑肌细胞1~7天,实时定量聚合酶链反应及Western blot检测血管平滑肌细胞CD36 mRNA和蛋白的表达,油红O染色测定血管平滑肌细胞内脂质含量。结果正常状态下血管平滑肌细胞存在微弱的CD36表达。与5mmol/L葡萄糖组比较,血管平滑肌细胞CD36 mRNA和蛋白水平的表达随着葡萄糖浓度的增加和干预时间的延长而增加(P〈0.05)。同时,细胞内脂质含量随着CD36表达增加而逐渐增加。结论高糖能促进血管平滑肌细胞CD36的表达,并促进血管平滑肌细胞内脂质聚集,有助于肌源性泡沫细胞形成。
Aim To investigate the effect of high glucose on CD36 expression in human vascular smooth muscle cells (VSMC) andon VSMC-derivedfoamccllformation. Methods VSMC was incubated with different dose of glucose (5, 11, 20 and 30 mmol/L) for one to seven days. Real time PCR and Western blot were used to measure the expression of CD36 in glucose treated-VSMC; Oil red O staining was used to measure the insular lipid accumulation. Results CD36 was expressed in human VSMC. The expression of CD36 mRNA and protein was induced by glucose in a dose- and time-dependent manner. Correspondingly, lipid droplets in the cytoplasm of VSMC were detectable and were gradually increased. Conclusion High glucose enhances the expression of CD36 and intracellular lipid droplets accumulation in VSMC, then inducing VSMC- derived foam cell formation.
