银杏胚愈伤组织的继代培养及其生理特性研究 被引量:4

摘要 以银杏的胚为材料诱导愈伤组织产生,研究愈伤组织生长量及其培养过程中过氧化物酶(POD)活性和ATP含量的变化,结果表明,2,4-D与BA组合比NAA和KT有利于各种外植体愈伤组织的诱导,NAA与KT组合更利于愈伤的继代培养。未成熟胚子叶愈伤组织的生长曲线大致为“S”型,在含BA的培养基上,愈伤组织前期生长较快,而在后期(24-28d)鲜重有所下降,POD活性和ATP含量在20d时达到最大;在含KT培养基上,愈伤组织在24~28d期间仍有增长,其POD活性和ATP含量在28d时最大,且有继续上升的趋势。这表明POD活性和ATP含量与银杏愈伤组织的生长有一定的相关性。 The present paper studied on the effects of plant growth regulators on subculture and growth of Ginkgo biloba embryo callus, and POD activity and ATP content of callus were studied. Theresults indicated that: 2, 4--D with BA were excelled to NAA with KT in callus induction, whereas NAA with KT were good in callus subculture. Immature embryo callus growth curve was " S" curve. In the medium containing BA, Callus early growed faster, and at a later stage (24-28 day) fresh weight reduced, POD activity and ATP content of callus were maximum in 20 days. In the medium containing KT, Callus fresh weight still increased in the 24-28 day and its POD activity and ATP content were maximum in 28th day. This shows that ginkgo callus growth related to the POD activity and ATP content in the process of subculture.
