摘要
目的检测经不同浓度甲醛暴露后导致金鱼脑细胞的遗传损伤。方法用不同剂量甲醛(10,100和1000μmol·L-1)对金鱼脑细胞进行体内染毒处理3天,利用单细胞凝胶电泳技术(彗星实验)检测脑细胞的DNA损伤。结果吸入浓度为10,100和1000μmol·L-1的甲醛后,可造成脑细胞DNA断裂。结论甲醛可造成脑细胞DNA的断裂,随着甲醛浓度的增大,DNA断裂程度加剧。
Objective To explore the DNA damage of brain cells of goldfish after formaldehyde exposure. Methods The goldfish were treated with liquid formaldehyde at different concentrations (10μmol/L, 100 μmol/L and 1 000 μmol/L) for three days. Single cell gelelectropho resis technique ( comet assay) was used to test the DNA damage of the brain cells. Results Formaldehyde caused significant DNA strand break at the concentration of 10 μmol/L, 100 μmol/L and 1 000μmol/L. Conclusion Formaldehyde was a DNA strand breaker,when the concentration is higher, it may cause crossliakagent to the brain invivo.
