复制缺陷性型腺病毒AdCMV-VEGF促进静脉皮瓣成活的实验研究

摘要 目的探讨复制缺陷性型腺病毒AdCMV-VEGF对静脉皮瓣成活的影响。方法于SD大鼠下腹部设计以腹壁下静脉为蒂的单蒂静脉岛状皮瓣,皮瓣区域注入重组的复制缺陷性型腺病毒AdCMV-VEGF(5×1011pfu个病毒颗粒溶于1ml生理盐水中)、AdCMV-Gal和生理盐水,2周后观察皮瓣成活面积,并进行免疫组化、原位杂交、常规组织切片检测。结果AdCMV-VEGF治疗组皮瓣平均成活率为22.3%,成活率高于AdCMV-Gal组和生理盐水组,差异显著(p<0.01)。无一例游离移植复合组织成活。结论应用复制缺陷性型腺病毒介导的人VEGF cDNA可增加非生理性皮瓣(静脉皮瓣)的成活面积。 Objective To explore the effects of repilication-deficient adenovirus AdCMV-VEGF on survival of venous flaps. Methods Uni-pedicled venous island flap based on the inferior epigastric vein was designed on the hypogastric zone of Sprague-Dawley rats. AdCMV-VEGF (5×10^11 pfu), AdCMV-Gal reconstituted in 1 ml normal saline were injected in the planned skin flap area subcutaneously. All animals were evaluated on postoperatively. The percentage of the survival area of each flap was measured and the distal viable skin flap tissue was harvested for histological analysis, hybridization in situ and immunohistochemical assay. Results The mean survival rates of AdCMV-VEGF group (22.3 %) were significantly higher than the AdCMV-Gal (0%) group and control (0%) group (p〈0.01). Conclusion The results suggest that local delivery of gene for VEGF via AdCMV can improve the survival area of ischemic venous flaps.
