大花惠兰组培苗中叶色突变体的获得与DNA鉴定 被引量:8

摘要 从大花惠兰的组培苗中获得了白色条纹和黄色条纹两种叶色突变体,突变发生的总频率约为0.12%。利用100条UBC随机引物将它们与对照进行了RAPD分析和对比。引物P17或P31可在对照和黄色条纹突变体间检测出多态性,黄色条纹突变体具有大小分别约为995 bp或1320 bp的多态性片段;而引物P84或P86则可在对照和白色条纹突变体间揭示出多态性,白色条纹突变体具有大小分别约为1120 bp或1315 bp的特异性条带。RAPD分析结果表明这两种突变体的基因组DNA都发生了变化。 Two kinds of leaf color mutants with white stripes and yellow stripes were obtained in regenerated seedlings of Cymbidium hyhridum and total mutation frequency was about 0.12%. The mutants were compared and analyzed with the control(their parents)by RAPD using 100 UBC arbitrary primers. The polymorphism between the yellow stripe mutant and the control was detected by using primer P17 or P31 and the former mutant showed the polymorphic fragment with size of 995 bp or 1 320 bp respectively, while the polymorphism between the white stripe mutant and the control was indicated by using primer P84 or P86 and the former indicated a specific band with size of 1 120 bp or 1 315 bp,respectively. Results of RAPD analysis suggest that the genomic DNA of two kinds of mutants has changed.
