血管内皮生长因子-2578C/A基因多态性与非霍奇金淋巴瘤关联的研究 被引量:2

摘要 目的检测非霍奇金淋巴瘤(non-Hodgkin lymphoma,NHL)患者血管内皮生长因子(vascularendothelial growth factor,VEGF)-2578C/A基因多态性分布,探讨VEGF-2578C/A基因多态性与NHL关系。方法采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)方法检测NHL患者及正常对照者的VEGF-2578C/A多态性位点的基因型。结果VEGF-2578C/A基因多态性在NHL患者与对照组之间差异无统计学意义(P>0.05)。依据NHL患者临床参数进行分层分析,骨髓浸润组A等位基因低于无骨髓浸润组(20.3%vs 27.9%,P<0.01或<0.05)。骨髓浸润组携带C/A或C/A+A/A基因型者低于无骨髓浸润组(31.4%vs 43.4%,36.0%vs49.6%,均P<0.01)。结论VEGF-2578CA或CA+AA基因型NHL患者发生骨髓浸润的可能性较低;-2578 A等位基因与减少骨髓浸润相关。-2578C/A多态性位点的基因型检测可能对判断NHL患者疾病侵袭性具有指导意义。 Objective To determine the distribution of vascular endothelial growth factor(VEGF) gene -2578 C/A polymorphism in non-Hodgkin lymphoma(NHL), and discuss the association between VEGF gene -2578C/A polymorphism and NHL. Methods Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was used to detect the VEGF -2578C/A genotypes in NHL patients and unrelated healthy controls. Results There was no significant difference in the frequency of VEGF-2578C/A genotype between NHL patients and healthy controls (P 〉 0.05). The genotype distribution and allelic frequencies of NHL patients were analysed for their clinicopathologic parameters. The frequency of the A allele in bone marrow infiltration group was lower than that in non-bone marrow infiltration group,with statistically significant difference (20.3% vs 27.9%, P 〈0.01 or 〈0.05). The VEGF -2578C/A and C/A + A/A genotype frequency in bone marrow infiltration group were lower than those in non-bone marrow infiltration group(respectively 31.4% vs 43.4%, 36.0% vs 49.6%, both P 〈0.01). Conclusion The C/A and C/A +A/A genotype are associated with weaker bone marrow infiltration in NHL patients. A allele is associated with weaker bone marrow infiltration in NHL patients. Determination of the VEGF-2578C/A genotype may be used as a stratification marker to predicate high-risk individuals for NHL.
