摘要
以脱脂奶粉作为初筛底物,纤维蛋白作为复筛底物,透明圈作为筛选标记,从肉联厂,污水沟等地筛选出产纤溶酶的菌株40株,从中筛选出产酶活力最高的2号菌株,将其编号为ZLW-2,并采用改进的紫外分光光度法测酶活,提高了酶活测定的精确性,对提高纤溶酶产生菌的筛选效率有重要意义。菌株ZLW-2的发酵过程研究表明,纤溶酶是在菌体大量生长以后酶活力才开始增加。
Forty fibrinolytic enzyme-producing strains were isolated from shambles, sewage,farm and so on, using the soluble circle as screening marker. The basal medium with skim milk was used as the first screening medium and the fibrin powder was used as the second screening medium. Strain 2 which had the highest fibrinolytic enzyme was chosen and numbered ZLW - 2. Moreover, the method of measuring enzyme activity was renovated and the curve of enzyme production was primarily studied. Ultraviolet photometry was adopted to measure enzymatic activity in the experiment, which had improved accuracy of the measurement and had great significance for improving efficiency of screening strains producing fibrinolytic enzyme. The fermentation process of ZLW - 2 indicated that the activity of fibrinolytic enzyme increased after mass growth of thali.
