产纤溶酶菌株S-05的液体发酵条件优化 被引量:4

摘要 对分离获得的一株产纤溶酶能力较强的芽孢杆菌(Bacillussp.)S-05,利用单因素试验和正交试验确定该菌株产生纤溶酶的适宜发酵培养基为:葡萄糖1.0%,大豆蛋白胨1.0%,Na2HPO40.4%,NaH2PO40.2%,MgSO4.7H2O 0.05%,CaCl2.2H2O 0.01%;发酵条件为:接种量2%,培养基装液量30 mL(250 mL三角瓶),培养温度30℃,摇床转速150 r/min,发酵周期48 h,初始pH 7.0;在此条件下发酵液产酶活力(相当尿激酶活力单位)高达652.55 IU/mL。 Bacillus sp. S-05 with high fibrinolytic enzyme productivity was isolated from the soil. The fermentation medium and conditions of the strain were studied via single factor test and orthogonal design. The optimal fermentation medium was determined as follows: glucose 1.0 % , soypeptone 1.0 % , Na2HPO4 0.4 % , NaH2PO4 0.2 % , MgSO4·7H2O 0.05 % , CaCl2 · 2H2O 0.01%. And the best fermentation conditions were: inoculation volume 2%, medium volume 30 mL in 250 mL flask, temperature 30℃ , rotation speed 150 r/min, fermentation time 48 h and initial pH 7.0. Under such conditions, the activity of fibrinolytic enzyme (corresponding to the unit of urokinase activity) was about 652.55 IU/mL of fermentation liquor.
