摘要
目的探讨膜结合型前列腺素E合成酶1(mPGES-1)在前列腺癌(PCa)及前列腺增生(BPH)组织中的表达,并进一步分析其与环氧合酶2(COX-2)的相关性。方法采用免疫组化SP法检测30例PCa和20例BPH组织中mPGES-1和COX-2的表达,用图像分析软件分析阳性表达的灰度和面积,并采用直线相关方法分析mPGES-1和COX-2的相关性。结果BPH组织中mPGES-1和COX-2表达呈阴性(14/20)或弱阳性(6/20);PCa组织中mPGES-1和COX-2均呈现阳性表达(30/30)。在PCa中,mPGES-1和COX-2的阳性面积表达水平与Gleason评分呈正相关(r=0.633、0.607,P<0.01);mPGES-1与COX-2也呈正相关(r=0.959,P<0.01)。结论mPGES-1和COX-2在PCa中均为高表达,且与PCa的Gleason评分呈正相关;mPGES-1和COX-2的表达呈正相关,提示两者在PCa发生发展中共同发挥重要作用。
Objective To investigate the expression of membrane-associated PGE2 synthase (mPGES-1) and its correlation with cellular differentiation in benign prostatic hyperplasia (BPH) and primary prostate cancer (PCa), further analyze the relatiuonship between mPGES-1 and cyclooxygenase 2 (COX-2) in PCa. Methods Expressions of mPGES-1 and COX-2 were detected by immunohistochemical assay in 30 tissue samples of PCa and 20 samples of BPH. A linear correlation was applied to analyze the correlation between mPGES1 and COX-2. Results Negative (14/20) to weak expression (6/20) of mPGES-1 and COX-2 was found in BPH. In contrast, all of the cancer (30/30) tissue showed positive mPGES-1 and COX-2. The over expression of mPGES-1 and COX-2 was correlated with the increasing of Gleason grade in PCa (r=0. 633,0. 607;P〈0.01). The level of mPG- ES-1 positively correlated with the level of COX-2 (r=0. 959,P〈0.01). Conclusion The proteins of mPGES-1 and COX-2 are over expressed and the level of mPGES-1 and COX 2 expression is positively correlated with Gleason grade in PCa. The positive correlation between mPGES-1 and COX-2 suggests that they might play important roles in the tumor development.
