摘要 目的:研究神经干细胞向神经元定向分化过程中微管相关蛋白-tau的表达变化。方法:采用细胞培养术、免疫荧光术及免疫印迹法对神经干细胞向神经元定向分化过程中微管相关蛋白-tau表达变化进行观察。结果:在神经干细胞向神经元定向分化过程中,tau蛋白在分化初期以核周附近分布明显,随神经元的成熟均匀密集分布于胞质中及突起内,tau蛋白的表达量逐渐增加。结论:在神经干细胞向神经元定向分化过程中伴随有微管相关蛋白-tau的表达变化,随着神经元的成熟,构成细胞骨架,维持细胞形态。 Objective: To observe and study the expression of microtube-associated protein tau in neurorroriented differentiation for neural stem cells. Methods: Using the techniques of cell culture, immtmofluorescence and Western-blotting, we observed the expression of tan in neuron-oriented differentiation of neural stem cells. Results: The expression of tau changed during the different phases of the differentiation. In the early phase, it showed an obvious distribution around nuclei, and then intensively presented in the cytoplasm and neurite. Positive reaction of tau was extended into the neurites with the gradual growth of the neuron, and the expression of tau was increased significantly. Conclusion : Expression changes of tau are concomitant with the neurorroriented differentiation of neural stem cells, and with the maturation of neurons, which participte in the cell skeleton and sustain the cell shape.
