摘要
目的:研究足月分娩胎盘组织细胞的表型特征并探讨其意义。方法:采用流式细胞术检测人胎盘组织单个核细胞(MNC)中CD133+细胞及其亚群含量;免疫组织化学法分析胎盘组织CD133、CD34、KDR、vWF和CD144的表达分布。结果:流式细胞仪检测结果表明,在胎盘MNC中,CD133+细胞所占百分率为(1.7±1.1)%,其中CD133+CD34+细胞亚群的比例为(0.5±0.5)%;胎盘CD133+细胞中表达CD34的细胞占(24.1±11.2)%。免疫组织化学结果显示,绒毛间质存在CD133+和CD34+细胞;胎盘绒毛合体滋养层和细胞滋养层细胞也表达CD133;而KDR、vWF和CD144的表达限于胎盘绒毛毛细血管内皮细胞,其表达CD34,但不表达CD133。结论:人足月胎盘组织间质中存在CD34+和CD133+细胞,晚期胎盘绒毛血管无内皮祖细胞存在。
Objective: To study the phenotypic properties of cells in human mature placental tissue (PT) and explore its significance. Methods: The proportions of CD133^+ cells and their subpopulation in PT derived mononuclear cells (MNC) were analyzed by flow cytometry (FCM). The expressions and localizations of CD133, CD34, KDR, vWF and CD144 in placental tissue were examined with immunohistochemical technique. Results: FCM results showed that the proportions of CD133^+ cells and CD133^+ CD34^+ cells in placenta derived MNC were (1.7±1.10) % and ( 0. 5 ± 0. 5) %, respectively, and ( 24. 1 ± 11. 2) % of CD133^+ cells expressed CD34. Immunohistochemical results showed that there were CD133^+ cells and CD34^+ cells in villous mesenchynx Syncytiotmphoblast and cytotrophoblast cells also expressed CD133. KDR, vWF and CD144 only expressed in placental villous capillary endothelial cells, which expressed CD34 but not CD133. Conclusion: Human placenta mesenchym of full-term deliveries containes CD34^+ and CD133^+ cells. There is no endothelial progenitor cell in the villous capillary of the mature placenta.
