摘要
背景与目的:肿瘤是一类多步骤发生、多基因突变及基因表达水平改变所致的细胞克隆性进化性疾病,其最基本的特征是细胞失控性生长。研究发现p27可能以计量依赖方式抑制细胞周期素依赖性激酶(CDK)复合物,从而抑制细胞增殖;而生存素作为迄今发现的最强的凋亡抑制因子,通过与半胱氨酸天冬氨酸酶(caspase)直接或间接的作用来阻断凋亡,实现细胞的恶性增殖。本研究旨在探讨抑癌基因p27和凋亡抑制蛋白生存素在非小细胞肺癌(NSCLC)组织中的表达、意义及其相关性。方法:采用免疫组化法检测60例非小细胞肺癌组织和20例正常肺组织中p27、生存素基因的表达。结果:①p27基因在非小细胞肺癌组织中的阳性表达率为40.0%,与正常肺组织相比,差异有显著性(P<0.01),并随病理分级的增加、临床分期的进展和淋巴结的转移逐渐下降(P<0.05)。②survivin基因在非小细胞肺癌组织中的阳性表达率为63.3%,与正常肺组织相比,差异有显著性(P<0.01),并随病理分级的增加、临床分期的进展和淋巴结的转移递增(P<0.05)。③p27、生存素基因在非小细胞肺癌组织中的表达密切相关(P<0.01)。结论:p27基因的低表达和生存素基因的诱导在非小细胞肺癌的发生发展中起重要作用,联合检测可为非小细胞肺癌的早期诊断、进一步治疗及预后判断提供必要的理论依据。
Background and purpose: Tumor is a disease associated with multi-gene mutation and the abnormal expression of multi-gene expression in many steps through the process of the evolution of cell clone. A lot of oncogenes and anti-oncogenes like p27, survivin, etc. take part in the regulation of the cell cycle directly or indirectly. We studied the expression and correlation of p27 and survivin in NSCLC. Methods: The expression of p27 and survivin was detected in 60 NSCLC and 20 normal pulmonary tissues by the immohistochemical staining. Results: (1)P27 expression in NSCLC was 40.0%, significantly lower than normal puhnonary tissues(P〈0.01).With the increase of histological grade,clinic stage,lymph node status,the expression rate of p27 was decreased(P〈0.05).(2)Survivin expression in NSCLC was 63.3%, significantly higher than normal pulmonary tissues(P〈0.01).With the increase of histological grade,clinic stage,lymph node status,the expression rate of survivin was increased(P〈0.05).(3)The expression of p27 and survivin were significantly correlated in NSCLC(P〈0.01). Conclusions: p27 and survivin may play important roles in oncogenesis and progression of NSCLC. Detection of p27 and survivin protein may have a role in early diagnosis, treatment and prognosis of NSCLC.
