刺突蛋白亚单位基因疫苗在大鼠体内诱导的体液免疫反应

摘要 目的利用重组腺病毒表达SARS病毒蛋白N端,研究其在动物体内诱导的体液免疫反应,探讨将其进一步开发为SARS病毒基因疫苗的可行性。方法用截短的刺突蛋白S1亚单位(SS)腺病毒载体疫苗(Ad—SS)皮下免疫大鼠[1×10^7pfu/(鼠·次),每周1次,连续3周],用ELISA法测定血清中抗SARS—CoV IgG水平。结果Ad—SS在大鼠体内能诱导产生高滴度的抗SARS冠状病毒IgGs,IgGs在免疫开始2周出现,在最后一次免疫1~2周达到最高(最高滴度为1:2^8.5)。用Vero—E6细胞体外攻击保护试验检测免疫动物血清中SARS病毒中和抗体发现,Ad—SN免疫鼠血清能在体外保护Vero—E6细胞免受SARS病毒攻击,中和滴度达到1:2^6.9。结论Ad—SN能在大鼠体内诱导SARS病毒特异的保护性体液免疫,有望发展成为一种SARS基因疫苗。
