摘要
目的模拟临床治疗中机械应力对细胞的作用方式,研究连续应力作用于成纤维细胞的生长增殖效应。方法实验采用细胞机械牵拉培养模型和体外培养的3~5代的成纤维细胞,分为对照组(未牵拉组)、连续牵拉组和重复连续牵拉组,各组分别进行牵拉后细胞计数和MTT比色法检测。结果连续牵拉组的细胞数量和活性较对照组明显增加,重复连续牵拉组的细胞数量和活性较连续牵拉组明显增加。结论连续应力可以促进体外培养成纤维细胞生长增殖和活性增加.
Objective To simulate the stress of clinic treatment and to study the expression of the proliferation of fibroblasts treated with successional stress. Methods A cell culture model was designed which can load mechanical strain. The 3-5 pages fibroblasts were used and three groups were divideded as:blank control group (no stress group), successional stress group, repeated and successional stress group. The number of the cells of all the groups was counted, and the cell proliferation was tested with MTT. Results The number and vitality of the fibroblasts of the successional stress group increased compared with the blank control group, The number and vitality of the fibroblasts of the repeated and successional stress group increased compared with the successional stress group. Conclusion The experiment simulates the stress of clinic treatment very well,and proves that successional stress can result in the growth and proliferation of fibroblasts,and improve the vitality of fibroblasts.
