甲醛对凋亡相关基因Bcl-2 Bax在生精细胞表达的影响

摘要 目的研究长期在甲醛含量过高的室内环境中凋亡相关基因Bcl-2、Bax在睾丸和生精小管细胞中表达。方法利用医用甲醛制备大白鼠的甲醛毒性模型,采用免疫组织化学技术,检测Bcl-2、Bax蛋白在睾丸及生精小管中的表达。结果高浓度的甲醛作用于大白鼠2个生精周期,每个生精小管中Bcl-2蛋白表达的阳性细胞数目在高剂量组为27±2.47,低剂量组为35±4.61,均显著低于正常组93±2.81,(P<0.05)。每个细胞中Bcl-2蛋白表达强度(OD值)在高剂量组为0.132±0.024,低剂量组为0.113±0.021,均明显低于对照组0.183±0.031,P<0.05;Bax蛋白表达的阳性细胞数目在高剂量组为89±20.14,低剂量组为61±14.76,均显著高于正常组23±4.71,P<0.05。每个细胞中Bcl-2蛋白表达强度(OD值)在高剂量组为0.098±0.021,低剂量组为0.085±0.018,均明显高于对照组0.032±0.026,P<0.05。结论长期处于高浓度甲醛的室内环境中使睾丸和生精细胞中Bcl-2的表达降低,Bax的表达增强。
