摘要
通过改进MTT方法,以中国棉铃虫细胞系为供试细胞,建立起一种适应化学农药毒力测定的新方法.实验结果表明:改进后的方法灵敏度高,可读出每孔仅500细胞的OD值;溶解蓝紫色结晶物(甲)吸收峰在550~570nm之间,参考波长为590nm;测定昆虫细胞范围为每孔500~60000细胞,这一范围能满足细胞生测及基础研究中多种测定的需要.
An insect cell bioassay for pesticide was developed with an established cell line of Heliothis armigera by modified MTT method. It is found that the solvent of modified method, pH4.5,3% SDS isopropanol, is superior to the others. All samples were measured with ELISA scanner at wavelenth 560 nm for measurement and 690 nm for reference. The linearity can be obtained at insect cell numbers within the range of 500~60 000 cells per well.
