摘要
目的:研究高蛋白饮食对犬加替沙星药动学的影响。方法:8只Beagle犬随机交叉空腹或给予高蛋白饮食同时单次口服加替沙星胶囊17mg·kg-1,采用高效液相色谱法测定血药浓度,用3p97软件计算两者的药动学参数并比较评价。结果:空腹组和高蛋白饮食组的药-时曲线符合口服吸收一室摸型,主要药动学参数分别为t1/2ke(4.00±0.84)、(9.28±1.32)h;tmax(2.29±1.08)、(4.98±2.76)h;Cmax(6.06±0.87)、(3.42±1.20)μg·mL-1;AUC(0~t)(56.74±10.80)、(68.66±15.92)μg.h.mL-1,2组比较各参数均有显著性差异(P<0.05或P<0.01)。结论:高蛋白饮食明显影响犬加替沙星药动学参数。
OBJECTIVE: To study the effect of high protein diet on pharmacokinetics of gatifloxacin. METHODS: A randomized crossover design was performed in 8 Beagle dogs. In the two study periods, a single oral 17 mg · kg^-1 dose was administered to empty stomach or with high protein diet in dogs. Plasma concentration of gatifloxacin were measured by HPLC. The pharmacokinetic parameters were calculated by 3p97. RESULTS: The concentrationtime curves were a one compartment model in empty stomach or with high protein diet in beagle dogs. The t 1/2ke were(4.00± 0.84) and (9.28 ± 1.32) h, tmax(2.29±1.08) and (4.98±2.76) h, C (6.06±0.87) and (3.42±1.20) μg·mL^-1, AUC(0-t) (56.74±10.80) and (68.66± 15.92) μg · h · mL^-1, respectively. There was significant difference between the two groups in the main pharma- cokinetic parameters(P 〈0.05 or P 〈 0.01). CONCLUSION : The high protein diet can influence pharmacokinetic parameters of gatifloxacin.
