缺氧诱导因子-1α及碱性成纤维细胞生长因子在人乳腺癌组织中的表达及其与血管生成的关系 被引量:2

摘要 目的:探讨缺氧诱导因子-1α(HIF-1α)和碱性成纤维细胞生长因子(bFGF)在人乳腺癌组织中的表达,以及两者与乳腺癌血管生成的关系及作用机制。方法:用免疫组化检测67例乳腺癌及39例乳腺纤维腺瘤组织中HIF-1α和bFGF的表达;用CD105标记血管内皮细胞并计数肿瘤微血管密度(MVD)。结果:HIF-1α及bFGF在乳腺癌组织中的表达阳性率分别为80.6%(54/67)、73.1%(49/67),与乳腺纤维腺瘤组织中的表达阳性率[0.0%(0/39)、35.9%(14/39)]比有统计学意义(P<0.001);乳腺癌组织中HIF-1α与bFGF的表达呈正相关(rs=0.314,P<0.01);乳腺癌淋巴结转移阳性组中HIF-1α及bFGF的表达与阴性组比无统计学意义;HIF-1α及bFGF阳性肿瘤组织中的MVD均值显著高于阴性组织,且MVD随着HIF-1α及bFGF表达的增强而增加;淋巴结转移阳性组中MVD高于阴性组(P<0.05)。结论:HIF-1α和bFGF在人乳腺癌组织中均高表达,且两者具有明显的相关性;HIF-1α与bFGF间可能通过相互作用共同调节乳腺癌血管生成。 Objective: To investigate the expression of hypoxia-inducible factor 1 alpha(HIF-1α)and basic fibroblast growth factor( bFGF) in human breast cancer and their relationship with tumor angiogenesis. Methods: The expression of HIF-1α and bFGF was examined in 67 cases of breast cancer specimens, 39 cases of fibroadenoma of breast by immunohistochemical method and the microvascular density( MVD) was counted in specimens marked by CD105 in the vascular endothelial cells. Results: The expression of HIF-1α and bFGF was statistically significant between fibroadenoma of breast and breast cancer respectively( P 〈 0.001 ). The expression of HIF-1α was significantly correlated with bFGF in breast cancer( rs=0.314, P 〈 0.01). The expressions of HIF-1α and bFGF were no statistical significance in the positive and negative lymph node metastasis groups respectively. The MVD value in the cancer tissue of positive expression of HIF-1α and bFGF was significandy higher than in the negative expression, and the number of the MVD increased along with the high expression of HIF-1α and bFGF in cancer tissues. The value of MVD in the positive lymph node metastasis group was higher than in the negative group( P 〈0.05). Conclusion: There is obviously positive correlation between the expressions of HIF-1α and bFGF in breast cancer. The interaction of HIF-1α and bFGF may play an important role in the regulation of angiogenesis of breast cancer.
