吲哚美辛对视网膜色素上皮细胞增殖、DNA合成及凋亡的影响

Effect of Indomethacin on Proliferation,DNA Synthesis and Apoptosis of Cultured Human Fetal Retinal Pigment Epithelial Cells in vitro

摘要 目的探讨吲哚美辛(IN)对体外培养的人胚胎视网膜色素上皮(RPE)细胞增殖、DNA合成及凋亡的影响。方法分别采用MTT法和核酸蛋白分析仪测定IN对RPE细胞增殖及DNA合成的影响;用透射电镜和吖啶橙荧光染色对凋亡细胞作定性、定量分析。结果与空白对照组相比,IN处理组RPE细胞数量减少,DNA浓度降低,细胞凋亡数增多,差异均有显著性意义(均P<0.05)。结论IN能抑制RPE细胞增殖和DNA合成,并能诱导细胞凋亡;在一定范围内,凋亡细胞数量随IN浓度增加和作用时间延长而明显增多。 Objective To investigate the effects of indomethaein (IN) on proliferation, DNA synthesis and apoptosis of cultured human fetal retinal pigment epithelial cells (RPECs) in vitro. Methods RPECs were treated with different concentrations of IN. Proliferation of RPECs was measured by MTT assay. The amount of DNA was determined by nucleic acid protein analysis. The quantitative and the qualitative analysis of apoptotie RPECs was respectively performed by using an acridine orange (AO) staining method and transmission electron microscopy. Results Compared with those in the blank control group, proliferation was inhibited, DNA synthesis was reduced and the number of apoptosis cells was increased in RPECs of IN-treated group (all P〈0. 05). Conclusion IN could inhibit the proliferation and DNA synthesis and induce apoptosis of cultured RPECs in vitro in a dose- and time-dependent manner.
