Cx32基因在肝癌细胞株中的表达与甲基化调节作用

摘要 [目的]了解Cx32基因在肝癌细胞株(HepG2)中的表达及甲基化调节作用。[方法]采用不同药物浓度的甲基化转移酶抑制剂(5-杂氮脱氧胞苷5-Aza-CdR)分三组培养细胞株,免疫组化分析Cx32蛋白表达,甲基化特异性PCR检测Cx32基因的甲基化状态。[结果]肝癌细胞株Cx32蛋白表达减少,去甲基化后Cx32基因表达增加且定位异常,细胞生长减慢,三组细胞总阳性率分别为18.5%、77%、88%,具有显著性差异(P<0.01)。三组细胞Cx32基因甲基特异性PCR结果分别为甲基化、部分去甲基化及完全去甲基化。[结论]肝癌细胞株(HepG2株)中Cx32表达减少,受甲基化抑制,去甲基化可增加Cx32表达。
