摘要
目的从血糖、血清CRP及胰岛素抵抗和胰腺组织内细胞间黏附分子-1(intercellular adhesion molecule-1,ICAM-1)及转化生长因子-β1(transforming growth factorβ1,TGF-β1)等指标观察芪莲汤对2型糖尿病(type2diabe-tes mellitus,T2DM)大鼠的保护作用及部分机制。方法应用高糖高脂饲料联合链脲佐菌素建立大鼠糖尿病模型,模型成功后每日灌胃给予芪莲汤(高剂量15g.kg-1.d-1,低剂量7.5g.kg-1.d-1)连续6周。实验结束后测血糖,取血清测定胰岛素含量和C-反应蛋白(C-reactive protein,CRP),用免疫组织化学的方法检测胰腺组织中ICAM-1和TGF-β1蛋白表达。结果芪莲汤可显著降低2型糖尿病大鼠的血糖水平,给药组与模型组比较有显著性差别(P<0.05),血糖水平与药物剂量呈依赖关系,高剂量组和低剂量组比较有显著性差异(P<0.05)。两给药组与模型组比较胰岛素抵抗指数明显降低,统计有显著性差异(P<0.05),胰岛素抵抗指数有剂量依赖趋势,但高剂量组和低剂量组比较无统计学意义。芪莲汤能降低2型糖尿病大鼠血清CRP含量,两给药组与模型组比较有显著性差异(P<0.05)。免疫组化结果显示,给药组胰腺组织ICAM-1蛋白表达均低于模型组灰度值,比较有显著性差异(P<0.05),而给药组TGF-β1蛋白表达较模型组有升高,高剂量组与模型组比较有显著性差异(P<0.05)。结论芪莲汤对2型糖尿病大鼠有明显的保护作用,其保护机制可能与降低胰腺组织中ICAM-1及升高TGF-β1蛋白表达有关。
Objective To investigate the protection effect and the mechanisms of QiLian Soup on the type 2 diabetes mellitus (T2DM) rats. Methods Fifty SD rats were randomly divided into two groups: 40 rats treated with high calorie feeds and streptozotocin (STZ) were utilized to establish the model of T2DM rats; 10 rats were taken as normal group. Thirty successful T2DM rats were studied and randomly divided into 3 groups: model blank group, high dose group and low dose group. Model blank group and normal group were administrated with distilled water every day. High dose and low dose group were treated with QiLian Soup of 15g·kg^-1·d^-1 and 7. 5·kg^-1·d^-1 respectively for 6 weeks. The level of the fasting blood glucose (FBG), serum insulin and C-reactive protein (CRP) were measured. The expression of Intercellular adhesion molecule-1 (ICAM-1) and transforming growth factor β1 (TGF-β1) on pancreatic tissue were evaluated by immunohistochemical method. Results QiLian Soup can significantly decreased the levels of FBG and insulin resistance index (IR), compared with model blank group (P〈0. 05). The level of FBG depended on the dose of QiLian Soup and was higher in low dose group than high dose group (P〈0. 05). IR and serum CRP level and the expression of ICAM-1 in panereatic tissue were lower than those of the model blank group (P〈0.05). The expression of TGF-β1 in two drug groups was higher than that of the model group (P〈0. 05). Conclusion QiLian Soup can decrease the expression of ICAM-1 and increase the expression of TGF-β1 in pancreatic tissue,and to decrease the serum IR,FBG and CRP. QiLian Soup has significantly protected effect on T2DM rats.
