摘要
利用超高速冷冻离心和Sepharose 4B凝胶过滤两步法从日本血吸虫中间寄主-湖北钉螺血淋巴中提取获得一种含铜呼吸蛋白-钉螺血蓝蛋白.该血蓝蛋白具有典型的血蓝蛋白基本特征,含铜量为0.23%,在340nm处观测到该蛋白的紫外特征吸收峰.SDS-聚丙烯酰胺凝胶电泳(SDS-PAGE)检测该呼吸蛋白由两种蛋白质亚基组成,分子量分别为400KDa和250KDa.透射电子显微镜测定结果证实其四级结构为中凹的圆柱体,且以单体、二聚体和多聚体形式共存于血淋巴中.
A copper-dependent protein was isolated from Oncomelania hupensis by a procedure of ultracentrifugation and gel fdteration on Sepharose 4B column. The absorption spectrum of the protein showed there was characteristic band at 340nm in the UV and the copper content in this protein was about 0.23%, characterization of hemooyanin. Two bands were observed in the SDS - PAGE gel, one near 400KDa, another near 250KDa compared with the protein marker. Transmission electron microscopy suggested its quaternary structure adopted a large hollow cylinder formation, Decamers, didecameric and multidecameric structures were all observed.
