摘要
目的检测Polo-like激酶1(Polo-like kinase 1,PLK1)在人类3种前列腺癌细胞株中的表达。方法采用RT-PCR和Western Blot方法检测人类前列腺癌细胞株LNCaP,DU145和PC3中PLK1 mRNA和蛋白的表达。结果在LNCaP,DU145和PC3中PLK1 mRNA均呈高表达,其PLK1/-actin比值分别为1.34,1.31,1.37;PLK1蛋白的表达水平有差异,Western Blot结果显示PLK1/-actin比值分别为1.17,0.83,0.76。结论mRNA水平上,PLK1在3种前列腺癌细胞中均呈高表达;蛋白水平上,PLK1的表达在不同前列腺癌细胞中存在差异。
Objective To investigate the expression of PLK1 in human prostate cancer cells LNCaP, DU145 and PC3. Methods mRNA and protein levels of PLK1 in LNCaP, DU145 and PC3 cells were detected by reverse transcription-poly merase chain reaction (RT-PCR) and Western blot. Results mRNA level of PLK1 was 1.34 in LNCaP cells, 1.31 in DU145 cells, and 1.37 in PC3 cells, respectively. Protein level of PLK1 was 1.17 in LNCaP cells, 0.83 in DU145 cells, and 0.76 in PC3 cells, respectively. Conclusion At mRNA level, PLK1 is overexpressed in the three prostate cancer cell lines. At protein level, the expression is different in various prostate cance cells.
