Bag-1在大鼠缺血预处理肝胆管中的表达及意义

摘要 目的研究缺血预处理对大鼠缺血再灌注损伤肝内胆管上皮细胞凋亡及Bag-1表达及影响,探讨IP对IR损伤后胆管上皮细胞凋亡的保护作用及其可能机制。方法健康雄性SD大鼠30只,随机分为假手术组(SO组)、缺血再灌注组(IR组)、缺血预处理组(IP组),每组10只。制备IR及IP模型,取肝门周围组织行免疫组化、原位杂交染色检测Bag-1基因和蛋白的表达及行TUNEL染色检测胆管上皮细胞凋亡并计算凋亡指数(AI)。结果TUNEL染色显示胆管上皮细胞凋亡水平SO组最低、IR组最高、IP组居中,三组两两比较均有统计学意义。免疫组化染色及原位杂交染色Bag-1mRNA和蛋白在各组中的表达程度与TUNEL染色提示的凋亡程度相同,且两两比较有统计学意义。结论缺血预处理对胆管上皮细胞具有保护作用,可能与上调Bag-1mRNA和蛋白的表达有关。 Objective To explore the effect of ischemic preconditioning(IP) on apoptosis and expression of Bag-1 mRNA and protein in bile duct cells of ischemia-reperfusion(IR) injury. Methods Thirty SD rats were randomly divided into sham operation group(SO group,n=10),IR group(n=10) and IP group(n=10).The liver orthotropic ischemic reperfusion model was established according to Chien's method.TUNEL technique,hybridization in situ and immunohistochemistry were used to determine apoptotic index and the expression of Bag-1 mRNA and protein,respectively. Results Compared with SO group,apoptosis of biliary epithelial cell increased in IR group.Apoptosis of biliary epithelial cell was significantly lower in IP group than in IR group(P〈0.05).Compared with SO group,expression of Bag-1 mRNA and protein in biliary epithelial cell increased in IR group (P〈0.05) and IP group (P〈0.01). Conclusion The protective effect of ischemic preconditioning can be implemented by up-regulating the expression of Bag-1 mRNA and protein in biliary epithelial cell.
