摘要
目的筛选并分析猪囊尾蚴新基因,为猪囊尾蚴病患者的免疫学诊断提供特异的重组抗原及有效的疫苗候选分子。方法用猪囊尾蚴病患者血清抗体筛选猪囊尾蚴cDNA文库,将所获阳性克隆的插入片段进行扩增并测序,通过互联网NCBI Gen Bank和蛋白质分析软件进行分析。结果筛选阳性克隆,经分析具有738bp完整开放阅读框,是1个新基因,登录号EU221317,命名为Ts2。结论成功克隆了猪囊尾蚴Ts2基因片段,为进一步实验提供了条件。
Objective To screen and analyse the new genes of cysticercus cellulosae for providing the specific recombination antigen and efficient candidate vaccine. Methods The cysticercus cellulosac cDNA library was immunoscreened with pooled sera of the patients with Taenia solium neurocysticercosis. The inserted cDNA fragments from the positive were analysed through the internet NCBI GenBank software. Results A positive clone was obtained in immunoscreen of cysticercus cDNA Library and it was named Ts2. The sequence of Ts2 was one of the unknown genes (GenBank accession No. EU221317) in cysticercus. Ts2 had 738 bp-size complete open reading frame (ORF). Conclusion A new gene encoding protein of Taenia solium metacestodes is cloned into pMD18-T, which would provide the basis for further study,
