产黄青霉发酵过程中菌体量测定的研究 被引量:3

摘要 对产黄青霉(Penicillium chrysogenum)发酵过程中菌丝量的测定方法进行研究,提出了一种新的基于超声波破碎细胞以及二苯胺显色法进行菌体量测定的方法,此方法与常规的测定菌体干重的方法比较,能很好地反映发酵过程中菌体量的变化情况。该方法重现性好、准确度和精密度较高,用于含固形物的发酵液中菌丝量的测定,得到了满意的结果。 A method to measure the content of the mycelia in the fermentation process of Penicillium chrysogenum was studied. The method acquiring the mycelia content based on breaking the cell wall with ultrasonic and diphenylamine color reaction was put forward. Compared with traditional method which quantify the mycelia weight by drying the mycelia, the advantages of this method is: It can reflect the true situation of the mycelia in Penicillium chrysogenum fermentation. This method has good reproducibility, accuracy in rating and precision, can be used to measuring the mycelia content of the fermentation broth contained solid materials.
