热休克预处理供肝对肝移植大鼠细胞因子及γδT细胞的影响 被引量:1

EFFECT OF HEAT PRECONDITIONING OF RAT LIVER GRAFT ON THE CYTOKINE AND γδT OF RECIPIENTS

摘要 目的探讨热休克预处理供肝对肝移植大鼠术后细胞因子及γδT细胞的影响。方法建立大鼠原位肝移植模型,将大鼠分为对照组,热休克预处理组,移植术后3 d、5 d、7 d,12 d收集肝脏组织观察病理学改变,肝组织提取单核细胞流式检测γδT,取血行细胞因子及肝功能测定,并观察两组大鼠术后存活时间。结果对照组IL-2、IFN-γ高于热休克预处理组,IL-10两组间无差别,IL-4术后两组均不能测出。热休克预处理组γδT高于对照组。对照组大鼠中位生存期为9 d,热休克预处理组中位生存期为15天。结论热休克预处理可提高移植肝脏单核细胞中γδT的比率,延缓Th1细胞因子上升趋势,但对Th2细胞因子无明显影响,术后γδT比率和细胞因子分泌无相关性,热休克预处理可延长大鼠肝移植的存活时间。 Objective To study the effect of heat preconditioning of rat liver graft on the Cytokine, γδT of recipients. Methods The rats were randomly divided into two groups, control group and heat preconditioning group. Recipients were sacrificed on 3 d, 5 d, 7 d, 12 d after liver transplantation .and liver tissues and blood samples were collected. Recipients survival rate, histopathological were observed, Plasm levels of ALT, AST, TBIL were measured by automatic biochemical analyzers. IL-2, IFN-γ, IL-4 and IL-10 were assayed by ELISA, The γδT infiltrated in hepatic allograft was detected by Flow Cytometry. Results The IL-2, IFN- γ,concentration of control groups were higher than heat preconditioning group, There was no difference in IL-10 concentration between two groups. IL-4 was not detectable in two groups. The γδT ratios of heat preconditioning group was higher than control group. The MST of control group was 9 days and heat preconditioning group was 16 days. Conclusion Heat preconditioning elevated the ratios of rats in allograft mononuclear and delayed the ascensus of Th1 cytokine, but had not manifest effect on Th2 cytokine. There was no relation between cytokine and γδT after transplantation. Heat preconditioning prolonged the survived time of recipients.
