摘要
目的探讨2-甲氧基雌二醇(2-ME)对体外培养的U87胶质瘤细胞增生、凋亡及细胞周期的影响。方法将不同浓度的2-ME分别作用于U87细胞,采用倒置显微镜观察细胞形态学变化;四甲基偶氮哩蓝(MTT)法检测2-ME对U87细胞增生的抑制作用;流式细胞术分析2-ME对U87细胞凋亡及细胞周期的影响。结果1~15μmol/L浓度的2-ME在24、36、48及72h均可明显抑制U87细胞增生并诱导其凋亡,并在一定范围内呈时间和剂量依赖性;各亚组与对照组差异具有统计学意义(P〈0.05)。U87细胞经2-ME作用后G2/M期细胞增加,而G0/G1期和S期细胞减少。绪论2-ME可明显抑制U87胶质瘤细胞增生并诱导其凋亡,在一定范围内呈时间和剂量依赖性。
Objective To explore the effects of 2-methoxyestradiol (2-ME) on proliferation, apoptosis and cell cycle of U87 ceils in vitro. Methods U87 cells were treated with 2-ME of different concentrations and time in vitro, morphological changes of apoptotic cells were observed under microscope ; cell proliferation inhibition was detected by MTT colorimetric assay; cell cycle arrest and apoptosis rate were measured by flow cytometry (FCM). Results Exposure to 2-ME had a strong antiproliferative effect on human glioblastoma and caused an increase in the population of apoptotic cells, detected by flow cytometry in U87 cell line. A significant number of cells were blocked in the G2/M phases of the cell cycle. Concurrently, the population of cells in the G0/G1 and S phases decreased in U87 cell line. Conclusion The present study showed that in vitro 2-ME inhibits the growth of human glioblastoma U87 cell hne and induces apoptosis in a timedependent and dose-dependent manner.
