摘要
目的用小鼠巨细胞病毒(MCMV)Smith株建立BALB/c小鼠全身播散型感染模型。方法15只BALB/c小鼠随机分为3组,于腹腔内接种0.2 ml不同浓度小鼠巨细胞病毒悬液(分别为5×104、5×103,5×102 PFU);另5只设为正常对照组。各组分别于接种7 d后处死小鼠,取小鼠唾液腺分离病毒并观察小鼠主要脏器组织病理损害。结果从接种MCMV 5×103 PFU组小鼠唾液腺分离出小鼠巨细胞病毒,小鼠肝、肺、肾和心脏组织病理损害明显。结论建立小鼠巨细胞病毒全身播散型感染模型,可为抗巨细胞病毒药物筛选、疗效评估和疾病发病机制和疾病预后评估提供一个良好动物模型。
Objectives To establish a BALB/c mouse model of cytomegalovirus-induced systemic infection with murine cytomegalovirus (MCMV): Methods Fifteen BALB/c mice were randomly divided into three groups, and were infected with MCMV by intraperitoneal administration of different doses of MCMV (5 ×10^5, 5 ×10^3 and 5 ×10^2 PFU). Five BALB/c mice were used as the control group. All experimental mice were sacrificed 7 days after intraperitoneal injection. Tissues of each experimental mouse were removed under aseptic conditions. Salivary glands were taken for virus separation and histological changes in the main viscera and tissues were examined. Results MCMV viruses were detected in the salivary glands and histopathological changes were observed in the livers, kidneys, lungs and hearts. Conclusions The establishment of the BALB/c mouse model of cytomegalovirus-induced systemic infection makes it possible for screening of antiviral drugs, assessment of therapeutic effects and allows for evaluation of pathogenesis and prognosis of thedisease.
