甘珀酸对戊四氮点燃癫疒间大鼠海马GFAP和Cx43表达的影响 被引量:3

摘要 目的探讨甘珀酸(CBX)对戊四氮(PTZ)点燃大鼠星形胶质细胞及其缝隙连接的影响。方法30只SD雄性大鼠随机分为3组:对照组(NS组),点燃组(K组),点燃后干预组(CBX组),每组10只大鼠。NS组每日腹腔注射生理盐水。K组和CBX组大鼠每日腹腔注射PTZ 35 mg/kg至点燃,再分别腹腔注射生理盐水、CBX(10 mg/kg)干预3 d。观察三组大鼠惊厥行为的变化,以及免疫组化染色方法观察GFAP、Cx43在海马内的表达变化。结果①PTZ点燃癫间疒大鼠出现自发性抽搐,CBX抑制了点燃大鼠的自发性抽搐,各组自发性抽搐次数为K组>CBX组>NS组(P<0.05)。②点燃癫间疒大鼠GFAP-IR星形胶质细胞增多,胞体肥大,突起丰富,Cx43的表达增加;CBX干预后,GFAP-IR星形胶质细胞无明显改变(P>0.05),Cx43的表达减少(P<0.05)。结论CBX对点燃大鼠星形胶质细胞的增生活化无影响,但抑制Cx43的表达和癫间疒活动,提示胶质细胞及其缝隙连接在癫间疒的发生发展过程中具有重要作用,CBX有潜在的抗惊厥或协助抗惊厥的作用。 Objectives To investigate the effect of carbenoxolone on astrocytes and its gap junction of PTZ-kindled rats. Methods Thirty adult male rats were randomly divided into normal saline (NS) group, PTZ-kindled (K) group, and CBX group, each group with 10 rats. NS group rats received intraperitoneal injection of NS only. The CBX group and K group rats were first treated with PTZ (i.p 35 mg/kg) to establish the epilepsy model of kindled rats. After kindling, the rats in both groups received CBX (10 mg/kg, CBX group) or NS (K group) for three days. The difference of seizure frequency among three groups was analyzed. The expression of GFAP-IR and Cx43 in hippocampi of the rats was examined with immunohistochemistry method. Results Spontaneous seizures occurred in PTZ-kindled epileptic rats. The frequency of seizure was less in the rats of CBX group than those in the K group. GFAP-IR astrocyte increased significantly in hippocampi of the rats in K group. In addition, hypertrophy of the cell body, increase of cell protrusions and Cx43 expression were found. However, there were no significant changes in GFAP-IR astrocytes (P 〉 0.05) , and the expression of Cx43 was decreased (P 〉 0.05) in CBX group. Conclusions Carbenoxolone showed no effect in stimulating the proliferation of glias in PTZ-kindled rats, but it suppressed the Cx43 expression and epileptic activity. The study suggested that astrocyte and its gap junction played an important role in the development and aggravation of epilepsy. CBX might have potential an-tl-epileptic activity.
