大肠癌及癌旁组织中Bim、Bax、bcl-2的表达及意义 被引量:13

摘要 目的检测Bim、Bax和bcl-2在大肠癌及癌旁组织中的表达情况及其临床意义。方法选择98例大肠癌及相应距癌组织5 cm外的正常组织构建组织芯片,采用SP免疫组化法检测癌及癌旁组织中Bim、Bax、bcl-2基因的表达情况。结果Bim和Bax在大肠癌中的阳性表达率(28.57%,23.47%)明显低于癌旁(98.97%,98.97%)。bcl-2在癌组织的阳性表达率(79.80%)较癌旁(25.51%)高。bcl-2过度表达与大肠癌的浸润深度、淋巴结转移及临床分期有关;Bim表达下降或完全丧失与浸润深度、淋巴结转移及临床分期有关;Bax表达降低与组织分化、淋巴结转移及临床分期有关。相关分析表明Bim与Bax的表达呈正相关,与bcl-2的表达呈负相关。结论Bim、Bax与bcl-2在大肠癌的发生发展中起重要作用。Bim对bcl-2的抑凋亡功能有对抗作用,对Bax的促凋亡功能有协同作用。 Purpose To investigate the expression of Bim, Bax and bcl-2 in colorectal carcinomar and adjacent tissues and their significances. Methods The tissue microarray was constructed by 98 cases of colorectal carcinoma and the adjacent normal tissues 5 cm away from the tumor. SP immunohistochemical method was used to detect the expression of Bim, Bax and bcl-2 genes in cancerous tissue and adjacent normal tissue. Results The positive rates of Bim and Bax in colorectal carcinoma (28. 57% and 23.47% )were significantly lower than those in the adjacent normal tissue(98. 97% and 98.97% ). The positive rate of bcl-2 in cancerous tissue (79. 80% )was higher than that in the adjacent normal tissue(25. 51% ). The over-expression of bcl-2 was correlated to the invasive depth, lymph node metastasis and clinical staging; the absence or decreased expression of Bim was correlative with the invasive depth, lymph node metastasis and clinical staging, and the decreased expression of Bax was correlative with the differentiation, lymph node metastasis and clinical stage. The Spearman correlation analysis suggested that the expression of Bim and Bax was positively correlated, and that the expression of Bim and bcl-2 was negatively correlated. Conclusions Bim, Bax and bcl-2 may play important roles in the carcinogenesis and progression of colorectal carcinoma. Bim has an antagonistic effect to bcl-2, and has a synergistic effect to Bax.
