豚鼠变应性鼻炎模型血清组胺的测定 被引量:3

摘要 目的探讨组胺在变应性鼻炎发病机制中的作用。方法用橄榄油将甲苯-2,4-二异氰酸酯配成浓度为10%溶液作为致敏剂,滴鼻,建立豚鼠变应性鼻炎动物模型8例。另8例正常豚鼠作对照。致敏结束、模型成功后取血清,用双抗夹心ELASA法测定血清组胺的含量。结果模型组血清组胺含量为(9.59±1.11)ng/mL,高于对照组的(7.62±0.96)ng/mL(P<0.01)。结论甲苯-2,4-二异氰酸酯的橄榄油溶剂滴鼻可诱发豚鼠产生变应性鼻炎的症状和血清组胺的浓度升高,成功建立了变应性鼻炎的豚鼠动物模型。 Objective To investigate the role of blood histamine in the pathogenesis of allergic rhinitis. Methods Toluene 2,4- diisocyanate was mixed with Florence oil in a concentration of 10%, and then this solution was dropped into the nostrils of animals to induce sensitization. 8 guinea pig models were prepared as allergic rhinitis and 8 normal animals served as the blank controis. The content of blood histamine was determined by the ELASA method after the model was successfully established. Results The content of blood histamine of the model group was statistically higher than that of the control group [(9.59 ± 1. 11)ng/mL vs (7.62±0.96)ng/mL, P 〈0.01]. Conclusion Allergic rhinitis was successfldly established in guinea pigs by Toluene 2,4- diisocyanate combined with Florence oil. Blood histamine may play an important role in the pathogenesis of allergic inflammation.
