摘要
以多糖得率为指标,研究了六种蛋白酶酶解鲍鱼脏器获得多糖的效果,结果表明碱性蛋白酶为最佳用酶,并对其工艺中温度、pH、水解时间等影响因素进行了实验分析,确定最适条件为:温度45℃,底物浓度2.5%,pH10.0,加酶量2.0%,水解时间3h,在此条件下,鲍鱼脏器多糖得率为6.50%。
The extraction technique of the polysaccharide from abalone harslet was studied. Six kinds of proteinase were applied to assist the polysaccharide extraction. The most suitable one was found to be the alkaline proteinase. Main factors during the extraction process were investigated. The results showed that the temperature and pH optima were 45℃ and 10.0,respectively. The optimal concentration of the substrate and alkaline proteinase were 2.5% and 2.0% ,respectively. The optimum hydrolytic time was 3h. Under the above optimized conditions, the polysaccharide yield was 6.50%.
