碱性成纤维细胞生长因子对人牙周膜成纤维细胞在壳聚糖/磷酸三钙支架材料上生长的影响 被引量:3

摘要 目的:将碱性成纤维细胞生长因子(basicfibroblast growth factor,bFGF)、壳聚糖/磷酸三钙(chitosan/tricalcium phosphate,CTCP)支架材料及人牙周膜成纤维细胞(human periodontal ligament fibroblast,hPDLF)体外联合培养,以明确bFGF对hPDLF在CTCP复合材料上贴附、生长和生物学活性的影响及其浓度效应关系。方法:取第5代hPDLF接种在CTCP支架材料表面,bFGF实验组加入含有浓度分别为1、10和100ng/mL的bFGF培养液,对照组在普通培养基中培养。各组均在孵育24h及72h后对材料上的细胞进行计数。结果:与对照组相比,各浓度bFGF实验组培养24h后均可促进hPDLF在CTCP支架材料上的附着,差异具有统计学意义(P<0.05),其中10ng/mL为最有效浓度。同时培养72h后各浓度bFGF组均能促进hPDLF在CTCP支架材料上的增殖,差异具有统计学意义(P<0.05)。结论:hPDLF在CTCP支架材料上的附着和增殖可被bFGF所增强。 Objective To study the effects of basic fibroblast growth factor(bFGF) on human periodontal ligament fibroblast (hPDLF) growth on chitosan/tricalcium phosphate (CTCP). Method The growth of hPDLF on CTCP were studied using cell culture. Results Compared with controls, three groups of bFGF could stimulate the attachment of hPDLF on CTCP after 24 h (P 〈 0.05), with 10 ng/mL as the most effcective dose. After 72 h, three groups of bFGF also could promote the proliferation of hPDLF on CTCP (P 〈 0.05). Conclusion BFGF appeared to be a positive growth factor to promote the attachment and proliferation of hPDLF on CTCP.
