摘要
目的:探讨青鹏膏治疗大鼠胶原性关节炎的作用及初步机制。方法:制备Ⅱ型胶原诱导大鼠关节炎模型,采用足趾肿胀测量仪测定大鼠的关节肿胀指数,采用ELISA方法测定外周血IL-1β水平,采用免疫组化方法检测关节滑膜IL-1α及TNF-α的表达,并对关节滑膜的病理变化进行组织学观察。结果:青鹏膏对胶原性关节炎动物模型的足肿胀有明显的抑制作用,给药组在关节肿胀指数,关节腔IL-1α,TNF-α表达水平等方面比模型对照组有所降低。结论:青鹏膏具有改善胶原性关节炎症候的作用,该作用与其降低关节滑膜IL-1α及TNF-α表达有关。
Objective: To investigate the effect of Qingpeng paste (QP) on collagen-induced arthritis (CIA) in rats. Method: CIA was established in female Wistar rats with injection of type II bovine collagen at the base of the tail of animals. CIA rats were treated daily with external administration of different doses of QP or voharen beginning on the day after the onset of arthritis ( day 1 ) until day 20. Paw swelling rate and the serum levels of IL-Ifl were determined. Moreover, the expression of TNF-α and IL-α and histopathological changes in the arthritic joints were also observed. Result: QP markedly suppressed the paw swelling rate of arthritic rat, reduced the expression of TNF-α and IL-α in synovial membrane. Histopathological changes in the arthritic joints were also significantly ameliorated in the QP-treated versus vehicle-treated rats. However, the elevated serum levels of IL-1β in arthritic rats were not influenced by QP. Conclusion: The present findings demonstrate the protective property of QP on collagen-induced arthritis, mechanisms underlying it may be related to reduce the expression of IL-1α and TNF-α in synovial membrane.
