摘要
研究螺旋藻中藻胆蛋白、多糖和脂类等药用成分的综合提取新工艺。首先,通过细胞破碎和盐析提取出藻胆蛋白,得率为3.99%,纯度达到0.61。然后用70%的丙酮对藻渣中的脂类进行提取,得率为2.2%。最后提取多糖,应用正交设计得出实验的最佳因素水平为碱溶液浓度为0.1mol/L,提取时间为1h,提取温度为室温。经TCA除蛋白后,用苯酚-硫酸法测定多糖得率达4.02%,多糖纯度为83.2%。
This article researched on the comprehensive extraction method for the phycobiliprotein, polysaccharide, lipids and other pharmaceutical compositions in the Spirulina. First, phycobUiprotein was extracted by staving the algae cell and salting out. The yield of phycobiliprotein was 3.99% and the purity achieved 0.61. Second, lipids were abstracted with 70% acetone solution from the algae dregs. The yield of lipids was 2.2%. Finally, applying the orthogonal design experiment, best factor level of extraction polysaccharide were as follows: the alkaline solution density 0.1 mol/L, the extraction time 1 hour and the room temperature, After TCA removed the protein, the yield of the polysaccharide was 4.02% and the polysaccharide purity was 83.2% by the determination of phenol-sulfuric acid method.
