气味沙雷氏菌蛋白组分分离及其体外抗癌活性 被引量:1

Protein fractions isolated from bacteria serrtia oderifera and their tumor-inhibiting effect in vitro

摘要 用SDS-PAGE(聚丙稀酰胺凝胶电泳)对经超声波处理的气味沙雷氏菌(Serratiaodorifera)全菌体进行分离,切胶分别洗脱.经丙酮沉淀、脱盐处理后得到九个蛋白组分.将九个组分分别用L929,QGY肿瘤细胞为靶细胞进行细胞毒性试验(MTT),经初步鉴定其中两个组分具有抗癌细胞活性.组分F6相对分子质量范围为28000~33000.其抑制50%细胞生长的蛋白浓度(IC(50)):对L929细胞为0.04610mg/ml,对QGY细胞为0.01250mg/ml.活性组分F9相对分子质量范围在15500~17000之间,其IC(50)对L929细胞为0.02790mg/ml,对QGY细胞为0.04307mg/ml.而F6、F9对WISH细胞却没有抑制作用,表明F6、F9能够体外直接抑制癌细胞生长. Nine protein fractions were obtained from altrasonic treated Serratia oderifera. At first, the intact bacterial cells were isolated by preparating SDS polyacrylamid gel electrophoresis (SDS-PAGE),then the specific gel bands were collected and washed, finally the protein was precipitated with propanone and desalted. The protein product was subjected to priliminary characterization. The relative molecular mass of the protein fraction F, ranged from 28 000-33 000 and its 50% inhibition concentration (IC,,) to L929 and QGY cells was 0. 04610 mg/ml and 0. 012 50 mg/ml respectively. The relative molecular mass of F, ranged from 15 500~ 17 000 and its IG50 to L929 and QGY cells was 0. 027 90 mg/ml and 0. 043 07mg/ml respectively. Both F6 and F9 did not show inhibition effect on WISH strain cells.These results indicated that both F, and Fg could inhibit the growth of tumor cells directly in vitro.
