摘要
[目的]观察枳术汤对脾虚气滞便秘小鼠P物质(SP)和降钙素基因相关肽(CGRP)基因表达的影响,探讨其靶向调控机制。[方法]采用饮食、劳倦加泻下法复制脾虚气滞便秘小鼠模型,造模组分别给予枳术汤、麻仁软胶囊、0.85%氯化钠治疗及非药物干预(自然恢复),用RT-PCR法检测并比较各组小鼠胃肠道SP和CGRP mRNA水平。[结果]模型组小鼠SP mRNA水平较空白组降低,尤以胃窦降低明显;而CGRP mRNA较空白组升高,以胃窦升高明显。治疗后枳术汤组SP mRNA较其他组升高,而CGRP mRNA较其他组降低,差异有统计学意义(P<0.05)。[结论]枳术汤可提高脾虚气滞便秘小鼠胃窦SP基因的表达,降低胃窦CGRP的表达,从而促进胃肠动力,达到治疗便秘的目的。
[Objective]To observe the targeted regulation mechanism of Zhishu Decoction on the genes expression of substance P (SP)and calcitonin gene-related peptide (CGRP) in mice with spleen-deficiency constipation. [Methods]The mice model with spleen-deficiency constipation were established by exerting the factors of diets, tiredness and coprorrhea, and then the model mice were treated with Zhishu Decoction, MaRen elastic capsule, normal saline and non-drug treatment. The mRNA expression of SP and CGRP in the gastrointestinal tract were tested by RT- PCR. [Results] The SP mRNA in the model mice were significantly less than that of the normal ones, but the CGRP were higher, both of which were obvious in the sinus ventriculi. After treatment, the SP mRNA in the Zhishu Decoction group were significantly higher than that of the other groups, whereas the CGRP were lower(P〈0. 05). [Conclusion ]The Zhishu Decoction could increase the SP mRNA expression but decrease the CGRP mRNA, which maybe the targeted regulation mechanism of Zhishu Decoction to treat the spleen-deficiency constipation.
