食用仙人掌的组培快繁技术研究

Studies on Technique of Rapid Propagation for Edible Cactus via Tissue Culture

以食用仙人掌嫩茎切块为外植体进行组织培养,研究了6-BAI、AA和IBA对其组培快繁的影响。结果表明,诱导萌芽以在MS+6-BA1.0 mg/L+IAA0.3 mg/L培养基上效果最好,萌芽率达88.9%;最好的增殖培养基是MS+6-BA1.0 mg/L+IAA0.3 mg/L,增殖率为345.4%,且中下部分比上部增殖效果要好;在1/2MS+IBA0.5 mg/L生根培养基上,平均每个苗长2.67个根,且有的根长达5 cm。当苗高3 cm,根长2cm时,炼苗3 d后移栽于椰子壳∶珍珠岩为1∶1的混合基质中,成活率为95%。

Using the tender stems of Edible cactus as the explants,the effects of different medium on plant regeneration were examined.The results indicated that the appropriate medium for bud induction was MS＋6-BA1.0 mg/L＋IAA0.3 mg/L.the highest rate of bud induction was 88.9%.the medium of multiplication was MS＋6-BA1.0 mg/L＋IAA0.3 mg/L,the multiplication rate was 345.4%.the multiplication of the lower parts of the micro-cladode is higher.the medium of rooting was 1/2MS＋IBA0.5 mg/L,averagely every stem has 2.67 roots,some roots length was 5cm.When root length was over 2 cm,open lid for 3 days and transplant them into mixture medium（residue and perlite in equal proportion）with up to 95% survival.
