摘要
应用组织化学方法观察了小鼠体内旋毛虫囊包幼虫经丙硫咪唑作用后酶活性及糖原含量的变化,研究丙硫咪唑杀虫的生物化学机理。取感染小鼠后腿肌肉的囊包蚴进行RNA、DNA、SDH、LDH、ACP、ALP、ATPase及糖原的组织化学进行半定量测定;并对糖原、LDH、RNA进行显微分光光度计定量测定。研究结果显示:用药组RNA、DNA、SDH、ATPase、ACP活性以及糖原均低于对照组,而LDH活性则高于对照组,ALP的活性无显著性变化。表明丙硫咪唑可通过抑制旋毛虫幼虫的糖、核酸的代谢,以及ATP产生而起杀虫作用。
The enzyme activity and glycogen content in encysted larva of Trichinella spirals from mice treated with albenedaze (ADZ) were observed by using enzyme histochemistry methods.The encysted larva taken from leg muscle of infected mice was used to detect DNA,RNA,SDH,LDH,ACP,ALP,ATPase and glycogen semi quantitatively with histochemical methods.Glycogen,LDH,and RNA were also tested quantitatively with micro spectrophotometer. The results showed that the activity of DNA,RNA,SDH,ATPase,ACP and the content of glycogen were lower than those of controls,whereas the activity of LDH was high.There was no significant change in the activity of ALP.It is suggested that the mechanism of ADZ making encysted larva lethal is inhibiting metabolism of glycogen,RNA,and DNA,and ATP production.
