摘要
研究龙葵碱对肝癌细胞HepG2的诱导凋亡作用.采用倒置显微镜观察形态学变化,Annexin V-FITC联合PI双染法和Tunel法检测龙葵碱诱导人肝癌HepG2细胞凋亡作用.结果发现龙葵碱作用24 h后,贴壁细胞数量减少、体积缩小,变圆等明显凋亡形态;在激光共聚焦显微镜下观察到AnnexinV-FITC阳性信号为绿色荧光,PI为红色荧光;Tunel阳性信号为绿色荧光.表明龙葵碱通过诱导HepG2细胞凋亡达到抗肿瘤的作用.
To investigate the apoptosis effects of solanine in HepG2 cell. The morphology of cells is observed under transmission electron microscopy. Apoptosis of solanine is assessed with Annexin V - FITC/PI and Tunel. The result is that with the inverted microscope, after being treated with drug-solanine , cells grew against the wall and peeled off gradually, which showed the characters of cells apoptosis. With the laser confocal microscopic, the positive signal of Annexin V - FITC is green, and PI is red ; the positive signal of tunnel is green. Inhibition on tumor of solanine can be accomplished through inducing the apoptosis in HepG2 cells.
