摘要
目的评价4价Aβ1-15基因工程肽疫苗(4×Aβ1-15)接种成年恒河猴的安全性。方法4只雄性成年恒河猴均免疫接种4次,每次间隔2周,以肌肉注射法接受4×Aβ1-15接种,检查免疫前后恒河猴的血常规和肝肾功能指标;观察疫苗接种后恒河猴的脑、肝、脾、肾的组织结构。在整个实验过程中,常规检测恒河猴有无局部和全身反应。结果4价Aβ1-15接种后,恒河猴无局部和全身不良反应,血常规和肝肾功能指标无明显变化(P>0.05),肝、脾、肾未见组织结构的病理改变;脑内无出血表现。结论4价Aβ1-15基因工程肽疫苗(4×Aβ1-15)具有良好的安全性。
Objective To evaluate safety of multivalent Aβ1-15 peptide gene engineering vaccine(4 × Aβ1-15) in rhesus monkeys. Methods Four experimental adult rhesus monkeys were received intramuscular injections with 4 × Aβ1-15 vaccine for four times ,and the intermission was two weeks. The general examination of blood and hepatorenal function in rhesus monkeys were proceeded before and after vaccination. Pathological examination of brain, liver, spleen and kidney was observed. Systematic reaction and local reaction were observed during vaccination each time. Results There was no adverse local and systematic reaction appearing in the rhesus monkeys after vaccination with 4 × Aβ1-15. The results of determination of haematology and hepatorenal funcion showed that all data were in the normal range and had no distinct difference whenever before the first vaccination or after the fourth vaceination(P 〉 0.05 ). Furthermore, there were no pathological changes in microstructure of brain, liver, spleen, kidney of the rhesus monkeys with vaccination and there was non-hemorrhage in brain. Conclusion Multivalent Aβ1-15 peptide gene engineering vaccine (4 × Aβ1-15 ) has favourable safety.
