血管内皮生长因子-C在胃癌细胞株和胃癌组织中的表达及其临床意义

The role of vascular endothelial growth factor-C and its expression in the gastric cancer tissues and cell lines

目的探讨血管内皮生长因子-C(VEGF-C)在不同类型胃癌细胞株及组织中的表达及其与临床病理之间的关系。方法应用反转录-聚合酶链反应(RT-PCR)、Westernblot分别检测VEGF-CmRNA及其蛋白在胃癌细胞株中的表达,对53例胃癌及癌旁组织中VEGF-C的表达采用免疫组织化学SP法染色检测,Ⅷ因子抗体及Ⅳ型胶原抗体双重免疫组织化学染色(SABC)法进行微淋巴管染色及微淋巴管计数。结果在胃癌细胞株中均可检测到VEGF-CmR-NA及蛋白的表达,胃癌组织中,VEGF-C蛋白的表达与淋巴结转移、病理分期有关(P<0.05);淋巴管密度与VEGF-C蛋白的表达、淋巴结转移、病理分期有关(P<0.05)。结论VEGF-C表达与胃癌淋巴结转移密切相关,并有可能参与了胃癌淋巴管的形成。

Objective To study the relation between clinicopathological characteristic with the expression of vascular endothelial growth factor （VEGF-C） in different type of gastric cancer cell lines. Methods The expression of VEGF-C mRNA and protein in gastric carcinoma cell lines was examined by reverse transcription-polymerase chain reaction （RT-PCR） and Western blot respectively. Fifty and three cases＇ expression of VEGF-C in gastric cancer and in the para-cancer tissue were detected by immunohistochemical methods. Double immunohistostaining with Ⅷ factor antibody and Ⅳ collagen antibody was used to stain and count the micro-lymphatic. Results The expression of VEGF-C mRNA and its protein in all gastric carcinoma cell lines could be detected. Expression of VEGF-C in gastric cancer associated with lymphatic metastasis and pathological staging （ P 〈 0.05 ）. Lymphatic density also concerned with the expression of VEGF-C, lymphatic metastasis and pathological staging （ P 〈 0. 05 ）. Conclusion VEGF-C may play an important role in lymphatic metastasis of gastric cancer. , and could involve in formation of lymphangiogenesis.
